Polymorphisms of DAOA are associated with schizophrenia and bipolar disorder risk.
D-Amino Acid Oxidase Activator (DAOA) is found in peroxisomes where it triggers the degradation of the gliotransmitter D-serine, which is a strong activator of NMDA-type glutamate receptors. Increased activity of DAOA could reduce D-serine and NMDA receptor functioning. DAOA overlaps and shares a complementary chromosomal strand with G72. Polymorphisms of DAOA and G72 are associated with schizophrenia risk and diminished prefrontal and hippocampal function. A 2003 study by Hattori and colleagues also identified the DAOA/G72 as a strong candidate site for bipolar disorder although researchers have yet to identify a causal polymorphism.
